Comparing E4-positive elderly controls with E4-positive younger LOAD cases Lead Investigator: Alan Renton Institution : Mt Sinai E-Mail : alan.renton@mssm.edu Proposal ID : 587 Proposal Description: The APOE E4 allele is the strongest known genetic risk factor for late-onset Alzheimer???s disease (LOAD). However, a subset of E4-positive individuals reach old age without developing LOAD, suggesting that protective factors are involved. Using next generation sequencing data, we shall undertake a genome-wide association study, comparing E4-positive elderly controls with E4-positive younger LOAD cases. This ???phenotypic extreme??? study design will maximize our power to identify genetic variants protecting against disease in E4-positive individuals.
